Comparison of methods for mycobacteria isolation from swine feces BJM
Oliveira,Eugenia Márcia de Deus; Rodriguez,César Alejandro Rosales; Rocha,Vivianne Cambuí Mesquita; Ambrosio,Simone Rodriguez; Ohara,Patrícia Miyuki; Amaku,Marcos; Ferreira,Fernando; Dias,Ricardo Augusto; Leão,Sylvia Cardoso; Ferreira Neto,José Soares.
Swine mycobacteriosis is an important cause of carcass condemnation at abattoirs. One of the best ways to recognize the etiologic agent involved, in live animals, is the fecal isolation, as 94% of the lesions are located in the digestive tract. Therefore, the goal of the present study was to compare the performance of four decontamination methods followed by inoculation in three different culture media, totalizing twelve procedures of mycobacteria search from swine fecal samples experimentally contaminated. The swine feces were artificially contaminated with 0.02 g of Mycobacterium avium, PIG-B strain, and subjected to mycobacteria isolation trial. The protocols used were: 1) modified Petroff or basic method; 2) modified Lowenstein-Jensen or acidic method;...

Comparison of three decontamination methods for Mycobacterium bovis isolation BJM
Ambrosio,Simone Rodrigues; Oliveira,Eugenia Márcia de Deus; Rodriguez,Cesar Alejandro Rosales; Ferreira Neto,José Soares; Amaku,Marcos.
Sixty samples of tissue fragments with lesions suggestive of tuberculosis from bovine abattoirs, kept in saturated solution of sodium borate, were subjected to four treatments: 4% NaOH (Petroff Method), 12 % H2SO4 and 1.5% HPC (1-Hexadecylpyridinium Chloride) decontamination, and physiological saline solution (control). The HPC method showed the lowest contamination rate (3%) when compared to control (88%, p<0.001), NaOH (33%, p<0.001) and H2SO4 (21.7%, p<0.002). Regarding the isolation success, the HPC method was better (40%) than the control (3%, p<0.001), NaOH (13%, p=0.001) and H2SO4 (1.7%, p<0.001) methods. These results indicate that HPC is an alternative to the Petroff method.

Demographic characteristics of owned dogs and cats of Votorantim, São Paulo State, Brazil Ciência Rural
Baquero,Oswaldo Santos; Chiozzotto,Evelyn Nestori; Garcia,Rita de Cassia Maria; Amaku,Marcos; Ferreira,Fernando.
A two-stage cluster sample design was used to estimate population parameters of dogs and cats of the urban area of Votorantim, São Paulo State, Brazil, to support the planning and implementation of population management programs for companion animals. For dogs, the total and density estimates were 27,241 dogs (95% confidence interval [CI]=23,903-30,578) and 951 dogs km-2 (95% CI=835-1,068), respectively. For cats, these estimates were 5,579 cats (95% CI=3,595-7,562) and 195 cats km-2 (95% CI=126-264). Cost was the most prevalent reason for not sterilizing the animals (33%, 95% CI=23%-44%, Deff=6.1). The percentage of interviewees that expressed reasons for abandoning their animals was equal to 9 (95% CI=5-13, Deff=2.1). Other estimates were obtained,...

Fluorescence polarization assay, competitive enzyme-linked immunosorbent assay (ELISA-C) and indirect ELISA for the diagnosis of brucellosis in buffaloes (Bubalus bubalis) Ciência Rural
Paulin,Lília Márcia Silva; Samartino,Luis Ernesto; Conde,Sandra Beatriz; Federsoni,Igor Stefan Poppovic; Ferreira,Fernando; Amaku,Marcos; Ferreira Neto,José Soares.
The objective of the present study was to compare the performance of three serological tests for diagnosis of Brucella abortus infections in buffaloes (Bubalus bubalis). Serum samples collected from 696 adult females were submitted to the competitive enzyme-linked immunosorbent assay (ELISA-C), (I-ELISA), fluorescence polarization test (FPA), 2-mercaptoethanol test (2-ME) and complement fixation test (CFT). The gold standard was the combination of CFT and 2-ME, considering as positive the reactors in both CFT and 2-ME, and as negative those non-reactors. ROC analyses were done for C-ELISA, I-ELISA and FPA and the Kappa agreement index were also calculated. The best combinations of relative sensitivity (SEr) and relative specificity (SPr) and Kappa were...

Spatial clustering analysis of the foot-and-mouth disease outbreaks in Mato Grosso do Sul state, Brazil - 2005 Ciência Rural
Negreiros,Rísia Lopes; Amaku,Marcos; Dias,Ricardo Augusto; Ferreira,Fernando; Cavalléro,João Crisostomo Mauad; Ferreira Neto,José Soares.
In the southern region of Mato Grosso do Sul state, Brazil, a foot-and-mouth disease (FMD) epidemic started in September 2005. A total of 33 outbreaks were detected and 33,741 FMD-susceptible animals were slaughtered and destroyed. There were no reports of FMD cases in other species than bovines. Based on the data of this epidemic, it was carried out an analysis using the K-function and it was observed spatial clustering of outbreaks within a range of 25km. This observation may be related to the dynamics of foot-and-mouth disease spread and to the measures undertaken to control the disease dissemination. The control measures were effective once the disease did not spread to farms more than 47 km apart from the initial outbreaks.
